The VANTAstar-F Multimode Microplate Reader (BMG-Labtech)

The undergraduate researchers for this project will be trained to use the BMG VANTAstar-F multimode microplate reader to analyze TcGlcK activity and percent inhibition. We will also prepare IC50, Dixon, Lineweaver-Burk, and Michaelis-Menten plots. One goal of using the instrument is to understand the mode of inhibiton exhibited by this inhibitor scaffold, which is currently unknown. Acquiring Dixon plots is the best strategy for understanding if we have competitive, noncompetitive, uncompetitive, or mixed-mode inhibiton.

Undergraduate researcher Destiny O'Neill in the instrumentation laboratory. The VANTAstar-F microplate reader can operate in absorbance, fluorescence, and luminescence modes. Our TcGlcK assay is designed to use fluorescence by the production of NADPH.

About This Project

The etiological agent for Chagas' disease is the protozoan parasite Trypanosoma cruzi, which affects 6 - 7 million people primarily in Latin America. The chronic stage of infection causes cardiomyopathy and the enlargement of other organs resulting in disease burden and death. There is an urgent need to replace the clinical therapeutics (benznidazole & nifurtimox) and this proposal focuses on a compound screen to observe a hit expansion with the target T. cruzi glucokinase.
