We have DNA and PCR products!

For the past month, I have been extracting DNA from the 120 sawfish specimens I collected from Dr. Patricia Charvet's collection and we have some great DNA from these specimens! Overall the average DNA quantity for these specimens is 34 ng/ul which is higher than normal. This is great news and because the DNA quantity was so high I was able to PCR amplify 30 samples using some of my larger primer sets that are between 400-500 bp and some of my smaller primer sets closer to 200 bp. If a PCR gel has these glowing bands on the gel then it means that it has worked. You can see here on these PCR gels that I have PCR products! I will keep you all updated on how sequencing goes once I send some products in.

About This Project

The Critically Endangered Largetooth Sawfish is one of the most threatened of all marine fishes. They have historically been, and still are, exploited for their toothy saw. After global declines, Brazil is likely the last place this species is found in the Atlantic.This project aims to use DNA from old saws to assess Largetooth Sawfish genetic health in Brazil which is a critical step to giving this species a fighting chance against extinction.
